ACTH/Corticotropin Peptides

Corticotropin-releasing factor (CRF), a 41 amino acid-peptide widely distributed in the brain mediates endocrine as well as autonomic and behavioral responses to stress. Its receptor subtypes, CRF-R1 and CRF-R2, are expressed in the brain. CRF is hypersecreted from hypothalamic as well as from extrahypothalamic neurons in depression, resulting in hyperactivity of the hypothalamic-pituitary-adrenal (HPA) axis and elevations of cerebrospinal fluid concentrations of CRF. This increase in CRF neuronal activity is believed to mediate certain behavioral symptoms of depression involving sleep and appetite disturbances, reduced libido and psychomotor changes. The hyperactivity of CRF neuronal systems appears to be a state marker for depression because HPA axis hyperactivity normalizes following successful antidepressant treatment. The CRF system may play a significant role in the regulation of energy balance. CRF and CRF-related peptides, which elicit their effects through G-protein-coupled receptors known in mammals as CRF1 receptor and CRF2 receptor, are capable of strong anorectic and thermogenic effects. CRF, urocortin and urotensin I share amino acid sequences and they inhibit food intake in mammals.

Centrally administered N-terminal fragments of ACTH (1-10, 4-10, 4-9) display convulsant properties in rabbits.
ACTH (18-39) is known as the Corticotropin-like Intermediate Lobe Peptide. It stimulates insulin secretion as well as amylase and protein secretion in a dose-dependent manner similar to those of secretin and carbamylcholine.
Corticotropin-inhibiting peptide (CIP), the 7-38 fragment of human ACTH (1-39), is known to act as an antagonist of ACTH receptors. It does not have any corticosteroidogenic activity.
Sauvagine is a hypotensive and diuretic peptide. Originally isolated from the skin of the frog, Phyllomedusa sauvagei. It affects diuresis in the cardiovascular system and causes the release of ACTH and endorphins.
Urocortin III is a 38 amino acid peptide that is a member of the Corticotropin-Releasing Factor (CRF) family of peptides. Studies show that Urocortin III is highly selective for the Corticotropin-Releasing Factor 2 (CRF2) receptor and does not show affinity for the CRF binding protein
