RNA Polymerase II is the central component of the transcriptional machinery. The two largest components of RNA Pol II, RPB1 and RPB2, form the enzyme active site. The mAb 8WG16 recognizes the C-terminal heptapeptide repeat (YSPTSPS) present on the largest subunit of RNA Pol II (RPB1). Clone 8WG16 has been used to investigate the role of the C-terminal domain of the 22 kDa RNA Pol II subunit in the formation of the transcriptional complex. The antibody CTD4H8 recognizes the C-terminal repeat of the largest subunit of RNA polymerase II from HeLa and S. cerevisiae cells. This antibody recognizes both the phosphorylated and unphosphorylated forms of RNA polymerase II.
