    Fluorescence microplate reader: Capable of detecting emission at 590 nm with excitation at 545 nm.

Transglutaminases (TG) catalyzes irreversible post-translational modifications of proteins. This large family of enzymes contains at least 9 members including TG 1 to 7, the coagulation factor XIIIa and keratinocyte membrane bound TG form1,2. Under physiological conditions, TG doesn’t catalyze transamination reactions. After stimulation, by Ca2+ for example, activated TG catalyzes the cross-link of a glutaminyl residue with a lysyl residue. Additionally to this one, TG can use mono- and polyamines, -OH groups or water as co-substrate. Moreover, other reactions, important for cell viability are also catalyzed by TG. Because of their ability to promote protein aggregates, TG are probably involved in neurodegenerative diseases including Alzheimer’s disease, Huntington’s diseases, Parkinson’s disease or supranuclear palsy and are now considered as potential drug targets3-5.

SensoLyte® Transglutaminase Activity Assay Kit provides a convenient assay for the high throughput screening of TG modulators and inhibitors. Poly-D-lysine are coated on the plate surface. Biotin-TVQQEL peptide contained in the substrate will be covalently linked to lysine by TG. In the fluorimetric kit, the amount of immobilized biotin is determined using streptavidin-horseradish peroxidase (HRP), 10-Acetyl-3,7-dihydroxyphenoxazine (ADHP), and hydrogen peroxide (H2O2). HRP, in presence of H2O2, oxidizes non-fluorescent ADPH into the strong fluorescent resorufin. Otherwise, colorimetric Kit allows the quantification of immobilized biotin using HRP and TMB substrate. Resorufin fluorescence intensity and colorimetric signal generated by TMB are proportional to TG activity in the sample. Assays are performed in a 96-well microplate format.
