Quant-Peptides are customized peptides which are accurately quantified. They are intended to be used as standards for quantitative proteomics.
Classical quantitation methods such as Amino Acid Analysis are inherently variable, resulting in standards that are too inaccurate for high precision proteomic assays. Eurogentec has developed innovative technologies that guarantee the accurate quantification of peptide standards. Quant-peptides are available in two formats: without or with a Quant-Tag.

Both technologies offer high precision peptide standards, and may be complementary:
Quant-Peptides with no Quant-Tag* are absolutely quantified using a proprietary and highly optimised AAA-MS method. These are universal peptide standards which can be used in any proteomics application.
Quant-Peptides with a Quant-Tag** are absolutely quantified based on the inherent spectral properties of the proprietary tag. The Quant-Tag is coupled to the C-term of the peptide via an Arginine (R) or Lysine (K) residue and can be released by trypsin digestion. The precise molecular mass of the tag (1356,7 Da) can be used in assessing the cleavage efficacy, and hence in setting the optimal trypsinization conditions of a sample using i.e. MS:MS methods. To do so, the Quant-Peptide must be spiked into the sample prior to trypsin digestion. The Quant-Tag is then used as a reporter of the trypsin digestion efficacy : a peak corresponding to the tag mass is indicative of trypsin digestion.
Absolute quantification of protein-based biomarkers from complex samples is central to pharmaceutical and diagnostic proteomics. The identification and quantification of peptides derived from the protein of interest is routinely used in several mass spectrometry based methods, including Single/ Multiple Reaction Monitoring (SRM/MRM). Accurately quantified peptides is prerequisite to their use as internal calibrators.
In quantitative proteomic analyses, using identical amounts of peptide standard in each sample is mandatory, as this spiked peptide is the calibrator from which the peptide of interest, present in the sample as trace amount, is quantified. Therefore, Quant-Peptides are dispensed in a highly reproducible and controlled manner to ensure assay reproducibility and accuracy.
